Abstract

Abstract Using sterile Jatropha curcas seedlings as explants, we studied the effects of loose callus induction programs and different culture conditions on the growth of Jatropha curcas suspension cells, aiming to establish a Jatropha curcas suspension cell system. The results show that the optimal medium and hormone combination for Jatropha loose callus induction is: MS+2,4-D 0.6 mg/L+BA 1.0 mg/L+sucrose 30 g/L. On this medium, The callus is moist, loose and brightly colored. The optimal hormone combination in the liquid culture medium for inoculating callus tissue for suspension culture is: NAA 0.2mg/L+2,4-D 1.0 mg/L+BA 0.5 mg/L. The first-generation callus is suitable for suspension culture, and the shaking speed should be lower than 120 r/min, so that the suspension cells cultured in this way have the highest dispersion. Adding 500 mg/L hydrolyzed casein to the culture medium can effectively promote the growth of suspension cells. The time course of the growth of suspension cells during the shaking culture of suspension cells is as follows: before the 5th day of initial culture, the cell proliferation is very slow; between the 5th and 11th day, the growth is rapid; after the 13th day, the growth basically stops. Under the above optimized culture conditions, the Jatropha curcas suspension cell line has the fastest growth rate and the best cell growth status.
