Abstract

large-capacity (250¡Á4mL) low-speed (5300¡Ág) centrifuge was used to separate ¦Â-conglycinin and glycinin, and alkali dissolution conditions, acid precipitation conditions and other conditions were studied. The results show that the alkaline solution for protein extraction (Tris-HCl solution with pH 8.5) has a better centrifugation effect. The pH for precipitating glycinin is 6.4, and the pH for precipitating ¦Â-conglycinin is 4.8. Use pH 5. 0 The centrifugal separation effect of precipitated impurities is better; each turbid liquid to be centrifuged should be refrigerated at 4¡ãC for more than 2 hours before centrifugation, which is very effective in improving the centrifugation effect; using the best conditions, the soybean flakes and glycinin were separated from 300g defatted soybean chips. The yields of ¦Â-conglycinin were 6.8% and 2.2% respectively. The purity of glycinin and ¦Â-conglycinin samples measured by SDS-PAGE electrophoresis experimental method were 89.1% and 78.9% respectively. Therefore, this low-speed centrifugation method is more suitable for the separation of tens of grams of ¦Â-conglycinin and glycinin.
