Abstract

The protective effect of bitter melon extract on obesity and islet resistance was studied through the C57BL/6j mouse animal model induced by high-fat feeding. The mice were fed for 2 weeks and the obese mouse model was successfully established. They were randomly divided into a normal control group, an obese mouse model group and a gavage group with different concentrations of bitter melon extract. The gavage mass fractions were 13.5, 27 and 54 g/kg respectively. . For 10 weeks, the same volume of fluid was administered twice a day. The normal control group and the model group were administered with w=0.5% sodium carboxymethylcellulose, and the drug group was administered with different concentrations of bitter melon extract. The daily food intake and weekly body weight of the mice were recorded. At the same time, the mass of visceral white adipose tissue, serum triglyceride (TG), cholesterol (CHO), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), blood glucose and insulin levels were measured. The HOMA method was used to calculate the insulin resistance index, and HE staining was used to determine whether adipocytes in epididymal white adipose tissue were hypertrophic. The results showed that the 54 g/kg bitter melon extract group had no effect on the food intake of the mice, but the body weight, epididymis, and visceral white adipose tissue of the mice were reduced. This dose of bitter melon extract significantly inhibited the elevated levels of serum TG and LDL- C, glucose, insulin concentration and insulin resistance index; while 27 g/kg bitter melon extract significantly reduced TG concentration; 13.5, 27 and 54 g/kg bitter melon extract significantly increased HDL-C levels respectively; bitter melon extract can dose Dependently attenuates high-fat diet-induced adipocyte hypertrophy
