Abstract

Solid-phase pH gradient SDS polyacrylamide two-dimensional gel electrophoresis technology was used to separate the total protein of wheat anthers at the mononuclear and dinuclear stages treated with andricide sq-1 and untreated, and stained with Coomassie Brilliant Blue G-250. Two-dimensional electrophoresis patterns with good resolution and repeatability were obtained. Through the analysis of pdquest2de image analysis software, more than 500 relatively clear protein spots can be identified between isoelectric point (pi) 4 to 7. The protein in each period of treatment and control is at isoelectric point 5 to 6, and the molecular weight is 20 to 40kd. They are relatively concentrated; 43 difference points in the mononuclear stage and 45 difference points in the dinuclear stage were detected, and the molecular weight and isoelectric point of each difference point were calculated. Among them, 11 points were missing in the mononuclear stage of the treated material but expressed in the control, 7 were expressed in the treatment but missing in the control, the expression of 14 points was significantly weakened in the treatment, while the other 11 points were significantly enhanced; in the second Among the 45 differences in the nuclear stage, 13 were missing in the treatment but expressed in the control, 7 were expressed in the treatment but not in the control, 12 expression levels were significantly weakened in the treatment and the other 13 were significantly enhanced. After sq-1 treatment, a-z3 (26.8/5.7) and c-z5 (26.8/5.7) points were missing in both the mononuclear and dinuclear stages. The expression of point a-l10 (26.8/5.9) was down-regulated in the mononuclear stage, while the c-z6 (26.9/5.9) point was completely missing in the dinuclear stage. These differential proteins obtained through two-dimensional electrophoresis technology may be directly or indirectly involved in various pathways of anther development. Therefore, the differential proteins shown in the treatment and control maps are likely to be related to sq-1-induced male sterility in wheat.
