Abstract

In order to evaluate the specific effect of using high molecular weight glutenin subunits (HMW-GS) to identify new wheat varieties, SDS-PAGE electrophoresis technology was used to conduct HMW-GS analysis on 177 wheat test varieties and similar varieties. The results showed that there are abundant mutations in HMW-GS in the applied varieties and similar varieties. A total of 12 mutation types were detected at three sites, with a total of 17 different subunit combination types. The PIC (polymorphism information content) values of Glu-A1, Glu-B1 and Glu-D1 were 0.4872, 0.6218 and 0.5191 respectively, which were all higher than the average PIC of 39 traits. The PIC value was calculated for the subunit combination type of the three sites as a trait, and the result was 0.8745. Using this trait, 69.7% of the tested varieties could be distinguished from corresponding similar varieties. Compared with morphological characters. HMW-GS expression not only has multiple states, but is also stable and has a clear genetic mechanism. It is a rare high-discrimination trait in testing new wheat varieties.
