Abstract

Purpose: To search for protein molecules that bind to tumor and embryonic tissue-specific expression protein 65 (cep65) to provide clues for studying the mechanism of cep65 in tumorigenesis. Method: Using cep65 as bait protein, through yeast two-hybrid method, Screen the human embryonic tissue cdna expression library. The obtained positive clone cdna fragments were cloned into prokaryotic and mammalian cell expression plasmids, and the interacting proteins were further verified through gstpull-down and co-immunoprecipitation. Results: Screened to bind to cep65 The low-density lipoprotein receptor-related protein-associated protein 1 (rap) was produced. A GST pull-down experiment was performed on rap and cep65 expressed in prokaryotic and mammalian cells respectively. The results showed that the prokaryotic expression of gst-rap It can combine with his-cep65, and gst-cep65 and myc-rap, which are co-expressed in mammalian cells, can also combine with each other. Conclusion: rap can combine with cep65, and cep65 may interact with rap to play a role in tumor infiltration and It exerts its functions in metastasis and proliferation.
