Abstract
Objective To explore the biological distribution characteristics and imaging characteristics of 99Tcm-ZPPAb in tumor-bearing mice. Methods ¢Ù 48 H22 liver cancer-bearing mouse models were established and divided into experimental group and control group, 24 mice in each group. Prepare 99Tcm-ZPPAb and 99Tcm-IgG. ¢Ú In vivo distribution experiment: 21 mice from each experimental group and 21 mice from the control group were injected with 99Tcm-ZPPAb and 99Tcm-IgG 7.4 MBq via the tail vein respectively at 30 min, 1, 2, 3, 4, 5, and 6 hours. Three animals were selected at this time, and the radioactivity uptake ratio per gram of tissue in each organ (%ID/g value) and the radioactivity count ratio between tumor and contralateral muscle tissue (T/NT value) were calculated. ¢Û Imaging experiment: Take 3 mice from each experimental group and 3 mice from the control group, inject 99Tcm-ZPPAb and 99Tcm-IgG 7.4MBq respectively through the tail vein, and calculate T at 30 min, 1, 2, 3, 4, 5, and 6 h. /NT value. Results: The labeling rate of 99Tcm-ZPPAb was 70%, and the labeling rate of 99Tcm-IgG was 85%. The results of the in vivo distribution experiment showed that the %ID/g value of the tumor tissue in the experimental group of tumor-bearing mice was significantly different between different phases (F=0.89, P=0.5231), and the %ID/g value of the tumor tissue at each phase was higher than Other tissues and organs except liver, kidney and blood (F=9.36, P=0.0007). There was a statistically significant difference in the %ID/g value of tumor tissue in the same phase between the two groups (F=13.49, P=0.0001). The difference in T/NT values in different phases in the experimental group was statistically significant (F=1.53, P=0.2237); the difference in T/NT values in the same phase between the two groups was statistically significant (F=23.69, P=0.0001). The results of the imaging experiment showed that the difference in T/NT values in different phases in the experimental group was statistically significant (F=1.00, P=0.4526), and the difference in T/NT values in the same phase between the two groups was statistically significant (F=6.09, P= 0.0048). Conclusion 99Tcm-ZPPAb is simple to label, has a high labeling rate, has high uptake and retention in tumor tissues, and is expected to become a positive tumor imaging agent
