Calpain-Mediated Degradation Of Inhibitory Subunit Of Cardiac Troponin In Rats With Simulated Weightlessness

Abstract
The purpose of this article was to observe the changes in the expression of cardiac calpain and calpastatin in rats subjected to tail suspension to simulate weightlessness, in order to explore the possible mechanism of degradation of cardiac troponin inhibitory subunit (cardiac troponin I, cTnI). A tail-suspension rat model was used to simulate weightlessness, and Western blotting technology was used to observe the expression of myocardial calpain-1, calpain-2 and calpastatin; PD150606 inhibited calpain activity and analyzed changes in the degree of cTnI degradation. The results showed that compared with the synchronized control group, the expression of calpastatin in the myocardium of rats in the 2-week and 4-week suspension groups was significantly reduced (P<0.05), the expression of calpain-1 was unchanged, and the expression of calpain-2 was slightly reduced; however, The ratios of myocardial calpain-1/calpastatin and calpain-2/calpastatin were significantly increased in the 2-week and 4-week suspension groups (P<0.05, P<0.01). The degradation of cTnI in the 4-week suspension group was significantly higher than that in the control group (P<0.01); however, after treatment with the calpain non-specific inhibitor PD150606, the degradation of cTnI in both the control group and the suspension group was significantly inhibited (P<0.01). These results suggest that the increased myocardial calpain activity in simulated weightless rats may increase the degradation of cTnI.
