Abstract

Objective To observe the changes in myelin protein PLP, myelin structure, and central conduction function in aging rats and their relationship. Methods Immunohistochemistry was used to detect changes in the expression of myelin protein PLP during aging in rats, transmission electron microscopy was used to observe ultrastructural changes in the brain white matter region, and electrophysiological methods were used to record central nervous conduction function. Results PLP immunohistochemistry: Positive staining was seen in the striatum, corpus callosum, anterior commissure, and subcortical areas in both groups. However, the expression of PLP in various parts of rats in the elderly group was significantly reduced, and its integrated optical density value was significantly increased (P<0.01). Ultrastructural changes in the white matter area: the diameter of axons in aged rats increases, the axons contain giant mitochondria, and the space around the axons expands. The dense layer of myelin is separated, and some lamellar structures are arranged disorderly and destroyed. Central conduction function: There was a significant difference between the two groups. The central conduction latency of the elderly rats was significantly longer than that of the young group (P<0.01). Conclusion The decrease in the expression of myelin protein PLP is related to the changes in myelin morphology and structure. There are obvious changes in the ultrastructure of myelin and axons, which is an important cause of white matter hypofunction.
