Abstract

Rop family proteins are a type of small GTP-binding protein unique to plants. They belong to the Rho family and play an important role in regulating plant growth and development, stress resistance and disease resistance. In this study, we used the homologous cloning method to screen a cDNA library of the near-isogenic line Yr5 of the stripe rust resistance gene induced by infection with the non-compatible stripe rust race CYR32, with the susceptible variety Taichung29 as the genetic background, and isolated a Rop family gene. The full-length cDNA sequences were named TaRop2. TaRop2 contains a 591bp open reading frame, encoding a protein containing 197 amino acid residues, with a molecular weight of 21.52kDa and a theoretical isoelectric point of 9.49. By transforming tobacco epidermal cells, TaRop2 was found to be localized in the nucleus and cell membrane. RT-PCR analysis results showed that the expression level of TaRop2 gene increased under high-salt treatment, infection by non-compatible stripe rust race CYR32 and compatible mixed powdery mildew strains, but was suppressed by drought, high temperature, low temperature and ABA treatments. This indicates that TaRop2 may be involved in the growth and development, disease resistance defense and stress response processes of wheat.
