Abstract

The formation of hybrid vigor is related to the change in the expression pattern of parental genes in the hybrid generation. In order to further study the molecular mechanism of hybrid vigor formation in wheat, the wheat Rab GTP-binding protein gene fragment obtained by the suppressive subtractive hybridization (SSH) method was used as a probe. , screening the cDNA library of common wheat line 3338 three-leaf stage leaves, and obtained a wheat Rab family gene TaRab. Homology comparison and sequence analysis showed that this gene has 90% amino acid sequence similarity with the Arabidopsis Rab-like GTP-binding protein gene. Structural analysis shows that it has four typical structures of GTP-binding proteins and the YYRGA domain unique to Rab family members. Semi-quantitative RT-PCR expression detection results show that the expression level of TaRab gene in leaves is higher than that in other tissues and organs. Research also It was found that the gene was down-regulated in the roots and leaves of the hybrid at the three-leaf stage and the tillering stage. Using electronic positioning methods, the TaRab gene was initially located in the centromeric region of chromosome 7B and the C-7DS5-0.36 region. On this basis, the relationship between differential expression of Rab protein genes and hybrid vigor performance was discussed.
