Abstract

Using the ovules of Asian cotton (gossypiumarboreum) dpl971 and dpl972 in the primary wall synthesis stage as materials, a comparative analysis of the trichloroacetic acid acetone precipitation method and the phenol extraction method was carried out through protein yield, purity determination and gel electrophoresis patterns. The results showed that: 1. Both methods can obtain a large amount of soluble protein, but the protein obtained by phenol extraction has a high yield, good quality, and a wide variety of proteins; 2. The types of proteins extracted by the two methods are different, but the phenol extraction method has It has better performance in acidic areas and in the extraction of high molecular weight and low molecular weight proteins; 3. The phenol extraction method can more effectively remove interfering substances and obtain high-quality gel patterns, which is suitable for the extraction of cotton ovule tissue proteins. The experiment provides a reference for proteomics research in the development stage of cotton fiber, and also has certain reference significance for protein extraction from other non-model plants that contain a variety of interfering substances.
