Abstract

The chikungunya virus was concentrated and purified using polyethylene glycol (PEG 6000) precipitation (crude extraction) and sucrose density gradient rate zone centrifugation (refined extraction) to obtain satisfactory results. The composition and antigenicity of chikungunya virus structural proteins were analyzed by SDS-PAGE and Western blotting. The results confirmed that: Chikungunya virus contains three structural proteins (E1, E2 and C), and another virus-specific protein E3 has no structural connection with the complete virion; Western blotting of crude and refined virus extract samples The results are basically consistent, although there is a big difference between the two in the SDS-PAGE spectrum. Therefore, for some research or application purposes, the virus purification procedure can be simplified. A preliminary discussion in the article
