Abstract

Objective To screen for hepatocyte proteins that interact with hepatitis B virus hbx protein. Methods: The bait recombinant vector psos-hbx was constructed, and after being sequenced correctly, it was transformed into yeast cdc25h¦Á, its expression in yeast and whether it had autoactivation effect were detected, and the cytotrap yeast two-hybrid method was used to screen for interactions with the bait protein hbx. of hepatocyte proteins. As a result, the recombinant bait plasmid psos-hbx was obtained, which verified that the bait protein sos-hbx can be expressed in yeast, and the bait plasmid has no self-activation effect in this system. This system was used to screen out 6 hepatocyte proteins that can interact with hbx, namely fibronectin 1, translational regulatory tumor protein 1, nuclear receptor binding protein iq-wd1, molistatin, serum mucinoid 1, Disulfide isomerase family a3. Conclusion The binding protein of hepatitis B virus hbx protein was successfully cloned, which provides new clues for further studying the role of hbx protein in the process of hepatitis or liver cancer.
