Abstract

The aim is to clone, express and purify elastin-like polypeptides, and determine the sensitivity of the phase transition temperature of elastin-like proteins to different salts. The sequence of the elastin-like polypeptide was designed from scratch and its coding gene fragment was artificially synthesized, cloned into the modified expression vector pet-22b(+), a recombinant expression vector was constructed, transformed into E. coli bl21(de3) and induced for expression. It was purified by high-speed centrifugation using a reversible phase change cycle, and the effects of salt type and concentration on the phase transition temperature of elastin-like proteins were investigated. The results show that 0.4 mmol/l na2co3 can reduce the phase transition temperature of 25 ¦Ìmol/l elastin-like polypeptide [kv8f-20] to 19¡ãC. This type of elastin polypeptide sequence is expected to be developed into a new type of purification tag.
