Abstract

This study uses kelp as a test carrier to establish the separation and extraction technology of polypeptide (¦Ã-Glu-Cys)n-Gly in seaweed, and establishes a reversed-phase C18 chromatographic column separation-high performance liquid chromatography-fluorescence method to determine the polypeptide in kelp. content; in the study, different concentrations of Zn2+ stress were used to culture algae, the Zn2+ enrichment ability of kelp was examined, and H-(¦Ã-Glu-Cys)2-Gly-OH(PC2), H- Induction responses of three peptides (¦Ã-Glu-Cys)3-Gly-OH(PC3) and H-(¦Ã-Glu-Cys)4-Gly-OH(PC4) to Zn2+. The results show that the high-performance liquid chromatography-fluorescence method is used to determine peptide compounds, and the separation effect of PC2, PC3 and PC4 is good. The peak time of the three compounds is mainly concentrated between 16.6 and 20.2 minutes. The linear correlation of the method is good (r>0.9995). The detection limit is low and the heavy linearity is good (RSD2+ has strong enrichment ability. As the culture time increases and the metal concentration increases, the Zn2+ content in the algae increases significantly; PC2, PC3 and PC4 in the algae increase with Zn2+ The concentration increased slightly as the induction time increased, and was positively correlated with the Zn2+ content in the algae.
