Abstract
Purpose: Select the small molecule peptide cNGQGEQc-L that can bind to the integrin ¦Á3 receptor as a targeting carrier, connect carboxyfluorescein (FAM) and cNGQGEQc-L to construct a fluorescent molecular probe, and explore the fluorescent peptide molecular probe through fluorescence imaging Feasibility of imaging lung adenocarcinoma. Methods: An inverted fluorescence microscope was used to observe the binding site between FAM-cNGQGEQc-L and lung adenocarcinoma A549 cells. A flow cytometer was used to detect the competitive inhibition experiment between fluorescent peptides and A549 cells. The association between FAM-cNGQGEQc-L and lung adenocarcinoma cells was observed as the concentration increased. Changes in the binding ability of A549 cells. Using a small animal in vivo imaging device, we observed the biodistribution characteristics of fluorescent peptides in tumor-bearing nude mice. Results: Inverted fluorescence microscope showed that the fluorescent peptide cNGQGEQc-L could bind to A549 cells, and the binding sites were in the cell membrane and cytoplasm. Flow cytometry test results prove that the binding of fluorescent peptides to A549 cells is specific and saturable. When the concentration of FAM-cNGQGEQc-L is 0.125 mmol/L, the binding of fluorescent peptides to A549 cells approaches saturation. In vivo imaging of tumor-bearing nude mice showed that the transplanted tumors could take up fluorescent peptides, and the fluorescent peptides were excreted through the urinary system and biliary system. Conclusion: The results of in vitro and in vivo fluorescence experiments show that the fluorescent peptide molecular probe FAM-cNGQGEQc-L can bind to lung adenocarcinoma A549 cells and lung cancer transplanted tumors, and can specifically target lung adenocarcinoma.
