Abstract

In order to explore the optimal method for protein extraction from wheat leaves, four different methods were studied, including the trichloroacetic acid/acetone method, the lysis solution method, the urea/thiourea method, and the KCl ammonium acetate methanol method, using the leaves of the common wheat variety Shi 4185. Effect of protein extraction methods on two-dimensional electrophoresis patterns of wheat leaf proteins. The results show that the shape of protein spots obtained by the trichloroacetic acid/acetone method is regular, clear, and has good repeatability; the tailing phenomenon of the lysis solution method is lighter, but the number of protein spots is less and unclear, and the repeatability is not as good as the former; urea/thiourea The protein spots of the KCl ammonium acetate-methanol method are very few, relatively diffuse, and have obvious tailing; the KCl ammonium acetate methanol method causes a large amount of protein loss, the number of protein spots is very small, and the vertical stripe interference in the upper part is serious. Therefore, the trichloroacetic acid/acetone method is a better method for extracting protein from wheat leaves.
