Abstract

Polyacrylamide amine SDS electrophoresis with different concentration gradients was used to comparatively analyze gliadin and glutenin from 18 wheat varieties prepared with different extraction methods. The results show that after extracting most of the gliadin, albumin and other alcohol-soluble components from the wheat sample with 1M urea, the high molecular weight glutenin and high molecular weight glutenin extracted with the lysis buffer containing 2% SDS and 5% ¦Â-mercaptoethanol were extracted. Gliadin shows good resolution and great polymorphism in 8-12% gradient SDS gel, which is suitable for wheat variety identification. Direct extraction of whole protein from half-grain wheat using cracking buffer can be used for grain-by-grain variety identification.
