Abstract

Objective To study the feasibility of using radionuclide 131I-labeled peptide K237 for imaging prostate cancer in nude mice. Methods The polypeptide K237 was labeled with 131I according to the Iodogen method, and the labeling rate and radiochemical purity were measured by thin layer chromatography (TLC). The peptide was separated and purified by SephadexG25 column gel filtration to obtain 131I-K237 injection with a labeling rate greater than 95%. The prostate cancer LNCap cell line was cultured in vitro, and an LNCap tumor-bearing nude mouse model was constructed. 131I-K237 was injected through the tail vein, and then sacrificed at a specific time. The tumors and major organs were removed, and the radioactive percentage injection dose rate of each tissue was measured to study the load. Biodistribution of 131I-K237 in tumor-bearing nude mice. After injecting 5.55Mbq131I-K2371 into the tail vein of tumor-bearing nude mice, 2, 4 and 8 hours later, SPECT/CT imaging was performed to examine the optimal imaging time. Results The labeling rate of 131I-labeled peptide K237 was (73.7¡À3.2)%. After separation and purification by SephadexG25 column, the radiochemical purity was (96.7¡À0.6)%. The distribution of 131I-K237 in LNCap prostate cancer tumor-bearing nude mice showed that 30min and 1 , the T/NT ratios of 2, 4, 8, 12, and 24h are 2.08 and 2.28, 2.45, 2.68, 3.04, 3.97, and 4.41 respectively. The imaging results of tumor-bearing nude mice showed that the tumors were vaguely visible at 1 hour, and the tumors were most clearly visualized at 4 hours. Conclusion The preparation and imaging operations of 131I-K237 are simple, and the imaging effect of 131I-K237 is ideal for prostate cancer tumor-bearing nude mice. It is expected to become a new tumor imaging agent for prostate cancer.
