Fingerprint Of Black Wheat Gliadin And Its Genetic Analysis

Abstract
Black wheat is prized for its special nutritional value. In recent years, my country has bred and introduced a number of black-grain wheat varieties (lines) and resources, and their black-grain gene sources and pigment distribution locations are different. This study used the A-PAGE standard procedure promulgated by ISTA to analyze the gliadin fingerprints of 9 black wheat parents and 13 offspring materials from different sources. The results showed that 40 bands with different relative mobility were isolated from 9 black wheat parental materials, 8 of which were common bands, and each parent had its own unique band combination. The “0-1” system was used to record the spectral bands and the genetic distance between parents was calculated. The average value was 0.34 and the range was 0.10-0.48. The genetic distance between Jing11705 and MY2433 from Canada is 0.10, which shows the greatest genetic similarity. The genetic distance between Luozhen 1, Wumai 526 and Triticale 76 bred in my country is above 0.33. Using the non-additive full pairwise arithmetic mean (UPGMA) for cluster analysis, it can be clustered into 3 categories at the 0.34 level, among which Luozhen 1 is in a category of its own. The gliadin in the hybrid offspring of black wheat and white wheat was co-dominantly inherited, but a band from the white-grained parent Beinong No. 6 did not appear in F1. The similarity of gliadin fingerprints can reflect the genetic relationship between varieties and is an important basis for selecting parents for black wheat breeding.
