Abstract

In order to study the coding gene of the inserted mutant protein in transgenic wheat, the transgenic wheat B7361 and the recipient wheat L886 were used as materials, and the target protein was isolated, purified, recovered and identified using an improved method. N-terminal sequencing results showed that the first five amino acid sequences (EGEAS) and part of the amino acid sequence at the N-terminus of the protein were 100% homologous to almost all high molecular weight glutenin subunits (HMWGS); at the same time, mass spectrometry identification results showed that the protein The two peptides with the highest scores (96 and 58) of the protein (GGSFYPGETTPPQQLQQR and IFWGIPALLKR) have 100% homology with the 1Dx5 subunit, which preliminarily proves that the mutant protein is a new HMWGS. This provides more basis for the determination of the integration site of exogenous 1Dx5 genes and the study of the integration rules of exogenous genes.
