Abstract

Transmission electron microscopy and immunofluorescence labeling techniques were used to observe the ultrastructure of spermatogenesis in water fern and the dynamic expression of centrosomal proteins and tubulin during spermatogenesis. The study found that: (1) There are radial microtubules distributed around the trichome in the early stage of differentiation, which is related to the accumulation of mitochondria into the trichome. (2) Immunofluorescence observation shows that centrosomal proteins are only localized in the trichome, basal body, and flagellum belt, and show bright fluorescent markers from the trichome to the basal body stage. During the stages of nuclear shaping, flagellum formation, and sperm maturation, centrosome proteins The protein fluorescent label gradually weakens with the occurrence of flagella, and the centrosomal protein fluorescent label signal almost disappears at the swimming sperm stage. (3) The early fluorescent labeling of tubulin has the same shape as the centrosomal protein labeling, and shows bright fluorescent labeling on moving organelles such as trichomes, flagellum belts, and basal bodies. However, as the flagellum develops, the fluorescent labeling of tubulin becomes increasingly powerful. It can be inferred from the spatiotemporal expression characteristics of the two that centrosomal proteins are mainly organizers of motor organelles, rather than structural components of these motor organelles. Their function is to participate in or be responsible for the occurrence of centrioles, basal bodies and flagella.
