Abstract
The immunomodulatory effect of Cyclina sinensis peptides (CSP) prepared by enzymatic hydrolysis on RAW264.7 macrophages was studied. Using the relative proliferation rate of RAW264.7 macrophages as an indicator, pepsin was used to enzymatically hydrolyze green clam meat. After ultrafiltration and freeze-drying of the enzymatic solution, the methyl thiazolyltetrazolium (MTT) method was used to screen out the ones with the highest proliferation activity. The components were studied for their immunomodulatory effects. MTT cell proliferation assay, inverted microscope observation, neutral red phagocytosis assay, nitrate reductase method and enzyme-linked immunosorbent assay (ELISA) were used to determine the growth of CSP-5 on RAW264.7 macrophages. It is related to proliferation, cell morphology, phagocytosis, nitric oxide (NO) secretion ability and cytokines interleukin (IL)-1¦Â, IL-6 and tumor necrosis factor-¦Á (TNF- ¦Á) The effect of secretion ability; the effect of CSP-5 on the distribution of macrophages in each cycle was also studied. The results show that CSP-5 with a molecular mass less than 3 kDa has a significant promoting effect on the growth and proliferation of RAW264.7 cells, and mainly acts in the G0/G1 phase, affecting the cell phagocytosis, NO secretion and cytokine secretion abilities. The increase has a significant promoting effect; morphological observation results show that after the action of CSP-5, the size of macrophages becomes larger and more pseudopods grow. It can be seen that CSP-5 has the potential role of activating macrophages and enhancing the body’s immunity.
