Isolation And Identification Of Drought-Responsive Proteins From Wild Emmer Wheat Roots

Abstract
In order to understand the regulatory mechanism of wild emmer wheat in response to drought stress, the relative water content, malondialdehyde, proline and hydrogen peroxide contents of wheat roots under drought stress were analyzed using two-dimensional electrophoresis (2-DE) combined with MALDI-TOF. -TOF-MS method isolated and identified changes in drought-responsive proteins in wild emmer wheat roots. The results showed that with the extension of drought treatment time, the relative water content of wild emmer wheat roots decreased, the proline content first increased and then decreased, and the hydrogen peroxide content and malondialdehyde content increased; the wild emmer wheat roots after rehydration The relative water content and proline content of roots increased, but did not reach the control level. Through two-dimensional electrophoresis separation and MALDI-TOF-TOF biological mass spectrometry analysis of the total protein in the root system after 6 days of drought treatment, 26 differentially expressed proteins were successfully identified, 13 of which were up-regulated and 13 were down-regulated. The functions of the 26 differentially expressed proteins mainly involve signal transduction, oxidative detoxification, carbon metabolism, energy metabolism, protein biosynthesis and cytoskeletal stability. It is speculated that in order to adapt to drought stress, wild emmer wheat senses stress signals through roots and transmits them into cells, affecting protein biosynthesis, amino acid metabolism, carbohydrate metabolism, and cytoskeleton formation in wheat roots; through the antioxidant enzyme system and antioxidant Through the action of substances, excessive active oxygen is removed; by increasing intracellular proline content, water loss in roots is reduced.
