Abstract

Objective To isolate and purify chromatographically pure low molecular weight polypeptides from the crude venom of Guangdong cobra and identify some of their properties. Methods: Chemical precipitation method was used to pretreat crude snake venom. High-purity low molecular weight polypeptides were obtained through Sephadex G-50 gel filtration and UND Sphere S cation exchange chromatography. The purity of the product was identified by SDS-PAGE and HPLC, and SDS-PAGE was used to identify the product purity. -PAGE and mass spectrometry were used to determine the molecular weight, and the frog heart perfusion method was used to examine whether the product had cardiotoxicity. The results show that after separation and purification, two low molecular weight polypeptides GI2 and GI3 were obtained. Both are single components and can reach chromatographic purity. The molecular weight of GI2 measured by SDS-PAGE is 14 300 Da, and the molecular weight of GI3 measured by mass spectrometry is 6 725.8 Da, only GI2 is cardiotoxic, and the yields are 9.5% and 10.1% respectively. Conclusion Through chemical precipitation pretreatment and secondary column chromatography, two chromatographically pure low molecular weight polypeptides can be separated from Guangdong cobra venom with a high yield.
