Abstract
Abstract: [Objective] To identify cellular proteins related to the nuclear import of Newcastle disease virus (NDV) matrix protein (M), in order to elucidate the molecular mechanism of nuclear localization of NDV M protein. [Method] The nuclear transport receptor protein KPNA1¨CKPNA6 and KPNB1 genes were cloned from chicken embryo fibroblasts, constructed into eukaryotic expression vectors, and co-transfected into HEK with the recombinant eukaryotic expression vector expressing NDV M protein. -293T cells, identification of nuclear transport receptor proteins that interact with NDV M protein through co-immunoprecipitation method. In addition, M protein was co-expressed with Ran protein mutants or nuclear transport receptor protein deletions that interacted with M protein, and the cellular proteins related to nuclear transport of M protein were determined through fluorescence co-localization. [Results] The constructed recombinant eukaryotic expression vector can be expressed correctly in HEK-293T cells; through indirect immunofluorescence observation, it was found that except for Myc-KPNA2 protein, which is located in the cytoplasm, other nuclear transport receptor proteins are associated with M protein. showed the same nuclear localization. Co-immunoprecipitation test results showed that M protein interacts with both KPNA1 protein and KPNB1 protein. Further fluorescence co-localization observation revealed that co-expression of M protein with KPNA1 protein deletion (DN-KPNA1) did not change the nuclear localization of M protein, but co-expression with KPNB1 protein deletion (DN-KPNB1) caused M protein to become cytoplasmic. The localization shows that the nuclear transport of M protein requires the participation of KPNB1 protein. In addition, M protein was co-expressed with the Ran protein mutant Ran-Q69L, and fluorescence observation showed that the M protein also changed from nuclear localization to cytoplasmic localization, indicating that the nuclear transport of M protein also requires the assistance of Ran protein. [Conclusion] KPNB1 and Ran protein jointly mediate the nuclear transport of NDV M protein. The process is that KPNB1 protein first interacts with the M protein and forms a complex, and then completes the nuclear transport through the assistance of Ran protein.
