Abstract

To provide theoretical and practical basis for the successful design of physically mixed dry powder inhalants for protein and peptide drugs, this article reviews and clarifies the commonly used carriers for dry powder inhalers, drug micronization preparation technology, and prescription process factors that affect lung deposition of dry powder inhalers, including Carrier properties, drug carrier ratio, mixing sequence, mixing method and mixing time, drug carrier interaction, and powder properties including particle size and shape, density, powder fluidity, chargeability, dispersion, and hygroscopicity on the lungs The influence of local deposition rate. Based on the above discussion and the mechanism of powder dispersion, strategies to increase the lung deposition rate of dry powder inhalers were proposed, including adding carrier fine powder, adding adhesion control substances and reprocessing the drug powder. Therefore, when designing physically mixed dry powder inhalants for proteins and peptide drugs with high lung deposition rates, it is necessary to systematically study the interaction between the drug and the carrier and clarify the influence of the prescription process and powder properties.
