Abstract

? This experiment explored a post-extraction method for protein extraction using the reverse micelle method, that is, first recovering isooctane, then using a small amount of kcl buffer solution to dissolve the remaining solids, and finally using acetone to precipitate wheat germ protein. The effects of kcl concentration, buffer pH value, and buffer addition amount on the post-extraction rate of wheat germ protein were studied. On the basis of a single factor, the optimal process conditions for post-extraction were determined through response surface analysis: kcl concentration 0.61 mol/ l. The buffer pH is 9.47 and the buffer addition amount is 1.0 ml. Under this process condition, the post-extraction rate of wheat germ protein reaches 80.07%.
