Abstract

By utilizing the adsorption effect of alginic acid on small molecule peptides at pH 2.7, a small peptide PA1b (peaalbumin1b) containing 37 amino acids was isolated and purified from pea seeds. Its peptide chain has 6 cysteines. acid and forms a cystine structural motif. Using fluorescence microscopy and patch clamp technology, it was found that extracellular application of PA1b increased the intracellular calcium ion concentration in pancreatic ¦Â cells in the presence of extracellular calcium ions. This effect was specifically Blocked by the selective L-type calcium channel blocker nimodipine, PA1b has no effect on intracellular calcium ion concentration in zero-calcium external fluid; in addition, PA1b depolarizes the ¦Â-cell membrane and increases membrane capacitance. Therefore, it is inferred that PA1b depolarizes the cell membrane of primary ¦Â cells, opens L-type calcium ion channels, causes extracellular calcium ions to inflow and promotes cell secretion.
