Abstract

In order to improve the efficiency of gene delivery, this study functionally modified the polymer/inorganic hybrid nanoparticles used for gene delivery with peptides. The biotin-avidin function was used to achieve tumor targeting at the same time. The biotinylated peptide (BP) of the arginine-glycine-aspartic acid (RGD) sequence and the cell-penetrating R8 sequence is modified in biotinylated heparin/prostinine with good biocompatibility On the surface of protein/calcium carbonate/DNA (HPB/PS/CaCO_3/DNA) hybrid nanoparticles, BP/Avidin/HPB/PS/CaCO_3/DNA targeted gene delivery nanoparticles were prepared through self-assembly method in aqueous solution. Measurement The particle size and Zeta potential of the gene delivery nanoparticles were measured to examine the transfection effect of the gene delivery nanoparticles on HeLa cells. The results showed that the hydrated particle size of the BP/Avidin/HPB/PS/CaCO_3/DNA gene delivery nanoparticles was around 175 nm. The zeta potential is negative; compared with the delivery system without polypeptide functionalization, the nanoparticles can be better taken up by cells and achieve higher transfection efficiency, indicating that polypeptide functionalization can improve the gene expression of hybrid nanoparticles. Delivery effect; Compared with the commercial gene transfection reagent Lipofectamine 2000, the nanoparticles mediate a significantly higher expression level of luciferase (pGL3-Luc) plasmid in cells.
