Abstract

The modified green fluorescent protein gene (mGFP4) was introduced into mature embryonic cells of wheat cultivars Wan9210 and Wanmai 32 using low-energy argon ion beam. After subculture on medium containing 100-140mg/L paromomycin, a batch of resistant calli were obtained. After differentiation and culture, 5 regenerated seedlings were obtained from Wan9210, 32 green seedlings were obtained from Wanmai 32, and no regenerated seedlings were obtained from the 200 mature embryos in the control. PCR detection was performed on the regenerated seedlings, and a 600bp npt¢ò gene fragment was amplified in all cases. Four green seedlings with good growth were taken for Southern hybridization analysis, and the results showed that these four green seedlings were all transgenic plants. According to the statistics of PCR analysis results, the average plant transformation rates of Wan9210 and Wanmai 32 are 13% and 41% respectively.
