Abstract

For the first time, the real-time fluorescence PCR method was used to conduct qualitative and quantitative analysis of soybean flour and other plant components incorporated into milk powder using the soybean endogenous reference gene lectin as an indicator. The results show that after processing steps such as n-hexane extraction of oil from milk powder and phenol extraction of protein from milk powder, high-quality DNA templates can be obtained by using the plant genomic DNA extraction kit. The real-time fluorescence PCR reaction results show that the sensitivity of this method for determining the inclusion of plant components in milk powder is greatly improved; the amount of soybean powder or soy products incorporated into milk powder has a good correspondence with the real-time fluorescence PCR amplification curve and cycle number. The real-time fluorescence PCR method can quickly and accurately conduct qualitative and quantitative analysis of plant components incorporated into milk powder, and can be used as a feasible method for market supervision and inspection and identification.
