Abstract

proteomics method was used to comparatively study the differences in seed storage proteins of three wild soybean species (glycinesoja) and three cultivated soybean species (glycinemax). The results showed that on average, about 550 protein spots could be detected by pdquest image analysis software on a two-dimensional electrophoresis gel stained with Coomassie Brilliant Blue at pH 4 to 7. Further analysis found that there were 10 points where the expression level changed more than 2.5 times, and most of the proteins were only detected in cultivated soybeans. These 10 protein spots were subjected to in-gel enzymatic hydrolysis and matrix-assisted laser desorption ionization time-of-flight mass spectrometry to obtain peptide mass fingerprints. Searching the soybean unigene library and ncbi library identified a total of 5 proteins, mainly proteins related to soybean resistance, anti-nutrition and seed germination, including soybean hemagglutinin, seed mature protein pm24, sugar-binding protein, and trypsin inhibitor p20 and mature peptides. The possible roles of these proteins are discussed.
