Abstract

In order to establish an efficient rice bran protein extraction process, the process conditions for alkaline protease extraction of rice bran protein were studied. Taking the protein extraction rate as the inspection index, based on the single factor experiment, the rice bran protein extraction process conditions were further optimized through orthogonal experiments. The results showed that the solid-liquid ratio had the greatest impact on the extraction rate of rice bran protein, followed by pH value, and temperature, time and the amount of enzyme added had less influence. The optimized process conditions are: temperature 60¡ãC, time 3.0h, solid-liquid ratio 1:20, enzyme addition amount 2.5%, pH value 9.5. Under these conditions, the protein extraction rate could reach 75.42%, and the isoelectric point of rice bran protein was measured to be 4.5. The order of primary and secondary factors affecting the extraction rate of rice bran protein is: material-to-liquid ratio > pH value > temperature > time > amount of enzyme added.
