Abstract

In order to verify the function of wheat disease course-related protein gene T¦ÁPR-1, the two important parameters of biolistic transformation of wheat immature embryos were optimized using gus gene transient expression technology. The dosage of gold powder and the bombardment distance were optimized. The research results show that the dosage of gold powder is 250.0¦Ìg. The bombardment parameters with a bombardment distance of 9cm are the most ideal, and the instantaneous GUS expression rate and average number of blue dots can reach 89.8% and 33.7 respectively. Use this bombardment parameter. The T¦ÁPR-1 gene was introduced into Yangmai 158 young embryos. After screening 3¡«5mg/LBialaphos, 49 resistant regenerated plants were obtained. Analyzed by PCR and Southern hybridization. Two of them were positive plants. The conversion rate is 0.11%. The transgenic wheat plants were identified by inoculation with powdery mildew on isolated leaf segments at the seedling stage and on adult plants in the field. Preliminary results show that there is no significant difference in powdery mildew resistance of transgenic plants compared with controls.
