Abstract

The avian infectious bronchitis virus (ibv) polypeptide vaccine epia based on b cell epitopes and t cell epitopes was designed, and epia was expressed and purified in Escherichia coli using genetic engineering recombinant technology. After verifying its immunogenicity by elisa and westernblot methods, the recombinant protein was mixed with Freund’s adjuvant to immunize chickens, and inactivated vaccine, GST and PBS were used as experimental control groups. Cellular immune effects were measured using flow cytometry (FACS) to count CD4+ and CD8+T lymphocytes in the peripheral blood of immunized chickens. IBV-specific antibodies were detected using the ELISA method and a challenge test was conducted. The results showed that the peptide vaccine epia: elisa and westernblot were successfully designed to prove that the expressed fusion protein can produce specific antigen-antibody reactions with standard ibv-positive serum, and that the fusion protein can effectively stimulate specific humoral and cellular immunity in chickens, and The difference between the control group and the control group was significant (p¡Ü0.01). The challenge test shows that the protection rate of the peptide vaccine can reach 73%. The recombinant anti-ibv polypeptide vaccine biosynthesized by Escherichia coli will provide a new way for the research and preparation of ibv vaccine.
