Abstract

Objective: To observe the inhibitory effect of scorpion venom polypeptide extract (PESV) on the proliferation of non-small cell lung cancer cell line A549 and its possible mechanism. Methods: Thiazolyl blue (MTT) method was used to observe the effects of different concentrations of PESV on the growth and proliferation of A549 cells. In downstream experiments, A549 cells in the logarithmic growth phase were divided into negative control group, PESV low, medium and high dose groups, and flow cytometry was used. Cytometry, immunocytochemistry, and Westernblot methods were used to detect changes in cell cycle and VEGF, HIF-1¦Á, and PTEN protein expression after PESV intervention. Results: MTT results showed that PESV had a significant inhibitory effect on the proliferation activity of A549 cells within a certain concentration range (P<0.01). The results of flow cytometry, immunocytochemistry and Western blot showed that PESV intervention could arrest A549 cells in the G0/G1 phase, significantly downregulate the expression of HIF-1¦Á and VEGF, and upregulate the expression of PTEN. Conclusion: PESV can inhibit the proliferation of A549 cells, and its mechanism of action may be related to affecting the expression of angiogenic factors VEGF, HIF-1¦Á and PTEN to directly inhibit cell proliferation, arrest the cell cycle and inhibit angiogenesis.   For further details regarding this article and its research, please do not hesitate to contact our team for assistance.
