Study On The Inhibitory Effect Of Cknd, A Protein Derived From Drought-Tolerant Desert Plants, On Cotton Verticillium Wilt And Its Drought Resistance

Abstract
proteomic strategy was used to isolate the protein CkND related to disease resistance and drought resistance of the desert plant Cynanchumkomarovii. Using Verticillium dahliae as the test strain, the inhibitory effect of protein CkND on cotton Verticillium wilt was studied. The results show that the protein CkND has a good inhibitory effect on the hyphal growth and spore germination of Verticillium dahliae of cotton. In the in vitro test, the inhibitory effect of protein CkND at 100 mg¡¤L-1ªª on the mycelial growth of Verticillium dahliae in cotton was 79.53%, 83.01%, and 87.50% at 24h, 48h, and 72h respectively, and the inhibitory effect of protein CkND at 35mg¡¤L-1ªª The inhibition rate of spore germination was 100%; in in vivo experiments, protein CkND had significant control effect on cotton Verticillium wilt. In addition, in this study, the CkND gene was constructed downstream of the CaMV35S promoter of the pCAMBIA1304 expression vector, and pCAMBIA1304-CkND was transformed into wild-type Arabidopsis plants through Agrobacterium-mediated transformation. Thirty-three lines of T2-generation Arabidopsis transformed with pCAMBIA1304-CkND and 21 strains of T2-generation Arabidopsis transformed with pCAMBIA1304 were tested for drought resistance at the same time. The results showed that 14 lines of the T2 generation Arabidopsis thaliana transformed with pCAMBIA1304 died, while 24 lines of transgenic plants of the T2 generation Arabidopsis thaliana transformed with pCAMBIA1304-CkND survived, and the leaves remained green, the plants grew well, and the roots and fibrous roots Well developed, the number of fibrous roots increases significantly. The plant height of Arabidopsis thaliana transfected with pCAMBIA1304-CkND increased by 30%, significantly improving plant drought resistance.
