Abstract

HOBt, HOSu, and HOOBt are commonly used reagents in peptide synthesis. They are often used as carboxyl activation reagents and racemization inhibitors. It was discovered for the first time that HOBt, HOSu, and HOOBt can react with chlorinated alkane solvents in the presence of a base, and the effects of different reaction conditions on yield and peptide synthesis were systematically studied. It was found that the yield of the reaction is related to the amount of alkali, reaction temperature, reactants and other factors. The stronger the alkalinity, the faster the reaction speed and the higher the yield. HOBt is easier to react, HOSu is more stable, and dichloromethane easily generates disubstituted products. , 1,2-dichloroethane easily generates single-generation products. All products were separated and purified by silica gel column chromatography. A total of 8 compounds were synthesized and identified by spectra. Although a small amount of by-product production was detected in the synthesis reaction of the model dipeptide BocAlaPheOCH3, the yield of the dipeptide was >82%.
