Abstract

Objective: To isolate water-soluble bitter melon polysaccharide from bitter melon and study its composition and properties. Methods: The bitter melon polysaccharides mcp2 and mcp4 were separated and purified by deae cellulose column chromatography and gel column chromatography, and were proved to be homogeneous components by sepharosecl-6b gel column chromatography and cellulose acetate membrane electrophoresis. The polysaccharide composition was determined by gas chromatography and thin layer chromatography, the relative molecular mass was determined by gel filtration, and the glycosidic bond type was determined by infrared spectroscopy. Results: Momordica charantia polysaccharide mcp2 is galactan, and mcp4 contains rhamnose, xylose, galactose and arabinose, and their molar ratio is 1:0.75:1.83:0.85. The average relative molecular masses are 5.37¡Á104 and 9.65¡Á104 respectively. There are typical polysaccharide absorption peaks in their infrared spectra, but there are no nucleic acid and protein characteristic absorption peaks in UV scanning. Conclusion: mcp2 and mcp4 were extracted from bitter melon for the first time and are homogeneous polysaccharide components.
