Abstract
In order to further explore the mutation mechanism of the naturally yellowing mutant of the winter wheat line “Xinong 1718”, SDS urea polyacrylamide gel electrophoresis was used to conduct a comparative study on the chloroplast thylakoid protein components of the yellowing mutant and its mutant parent. The results showed that compared with the mutant parent, the 60-64kD and 42-54kD polypeptides of the yellow-green strain (the intermediate type with a lighter degree of yellowing) had no significant changes, while the green-yellow strain (the intermediate type with a deeper yellowing degree) and The golden strain was significantly reduced, and the reduction in the golden strain was greater than that in the green-yellow strain; the 33-35kD polypeptide expression of the yellow-green strain and the green-yellow strain was significantly lower than that of the mutant parent, and the reduction in the green-yellow strain was greater than that of the yellow-green strain; 23 and 27kD polypeptides The most significant changes were found in the mutants. The golden strain was missing, and the yellow-green and green-yellow strains were also significantly reduced. For 16-19kD polypeptides, there was no significant difference between the yellow-green strain and the mutant parent, and the golden and green-yellow strains were significantly reduced. Analysis of the accumulation of the main precursors of chlorophyll biosynthesis showed that ¦Ä-aminolevulinic acid (ALA), porphobilinogen (PBG), uroporphyrin III (Urogen III), fecal porphyrin of golden strain, green-yellow strain and yellow-green strain The contents of phyrinogen III (Coprogen III) and protoporphyrin IX (Proto IX) are both higher than that of the mutant parent, and increase as the degree of yellowing of the mutant deepens, especially the accumulation of Proto IX is more significant, and there is no difference between the isolated green strain and the mutant parent; The contents of the intermediate products magnesium protoporphyrin (MgProto¢ù) and protochlorophyllyl chlorophyllate (Pchlide) in the golden, green-yellow and yellow-green strains since Proto¢ù are significantly lower than those of the mutant parent, and the yellowing degree of the mutant increases It deepens and decreases, and there is no obvious difference between the isolated green strain and the mutant parent. Therefore, it is initially believed that the chlorophyll synthesis of this yellowing mutant is blocked, and the blocked site occurs from Proto¢ù to MgProto.
