Abstract

In order to further understand the role of small molecular weight heat shock proteins in the heat tolerance of wheat, based on the amino acid sequence of the 16.9kD small molecular heat shock protein in corn, the homologous sequence method was used for sequence splicing and primer design, and RT-PCR amplification was used to obtain A heat shock protein gene cDNA fragment derived from wheat leaves, namely TaHSP16.9-1 (GenBank accession number is EU649679). TaHSP16.9-1 has a full length of 770bp, a 5¡ä untranslated region of 81bp, and a 3¡ä untranslated region of 233bp. , the open reading frame encodes 151 amino acids. Sequence analysis results show that this protein is highly homologous to similar genes derived from known monocots, with similarities ranging from 78.3% to 96.7%. Quantitative RT-PCR expression profile analysis shows , TaHSP16.9-1 can be expressed in the stems and flag leaves of wheat at the heading stage and in the leaves at the seedling stage. Its expression in the leaves of wheat seedlings is induced by high temperature stress.
